| Description: | The enzyme activity was assayed by using an ATP regenerative NADH consuming system. The reaction was started with adding ATP to the mixture containing enzyme, substrate, and inhibitor. The decrease of NADH was time-dependent, and the absorbance changed at 340 nm was monitored. The inhibition constant, Ki for inhibiting PKA and ROCK were obtained from dose-response titration curves. |
