| Description: | The assay measured the NADH-dependent catalysis of a DD-CoA substrate as a decrease in 340 nm absorbance resulting from conversion of NADH to NAD. The absorbance OD readings were taken on a microreader. The IC50 values were determined using at least eight concentrations, with each concentration assayed in triplicate under saturating substrate conditions. The values were calculated from plots of enzyme activity versus the log of the inhibitor concentration using the GraFit IC50 four-parameter fit software. |
