| Description: | The HDAC enzyme in vitro assay was based on a homogeneous fluorescence release assay. Purified recombinant HDAC enzymes were incubated with compounds diluted in various concentrations in assay buffer before fluorescent substrate was added. After incubation, additional trypsin incubation allowed the release of the fluorophore from the deacetylated substrate. The fluorescent signal was detected by fluorometer: 360 nm excitation, 470 nm emission, 435 nm cutoff. The IC50 values of the compounds were determined by analyzing dose-response inhibition curves from 6-21 independent experiments. |
