| | IKK Kinase Inhibition Assay |
| Description: | IKK kinase activity was assessed using a time-resolved fluorescence resonance energy transfer (TR-FRET) assay. The reaction was initiated by the addition of GST-IKBalpha substrate/ATP to the reaction buffer containing IKK kinase and test compounds. After incubation, detection reagent containing antiphosphoserine IKBalpha monoclonal antibody labelled with europium chelate, and an APC-labelled anti-GST antibody was added. The degree of phosphorylation of GST-IKBalpha was measured using a Packard Discovery plate reader (Perkin-Elmer) as a ratio of specific 665 nm energy transfer signal to reference europium 620 nm signal. |
