| | Topoisomerase IIα Relaxation Assay |
| Description: | Relaxation of negatively supercoiled plasmid DNA by human topoisomerase IIα was assayed in 20 µL of reaction buffer (10 mM Tris-HCl, pH 7.9, 150 mM NaCl, 0.1% BSA, 0.1 mM spermidine, 5% glycerol and ATP) containing 250 ng of supercoiledpHOT1 plasmid DNA and 1 unit of enzyme. After incubation at 37 °C for 30 min, the reactions were terminated and analysed by agarose gel electrophoresis. The ethidium bromide-stained gel was photographed over UV light for densitometry analysis. |
