| | Enzyme Assay and Determination of the Inhibition Constants IC50 |
| Description: | Inhibition of the matrix metalloproteinases MMPs has been determined by continuously monitoring the hydrolysis of the fluorescent substrate. The hydrolysis was followed by measuring the increase in relative fluorescence. The enzyme activity was measured after 10 min preincubation of the enzyme with varying concentrations of the inhibitor. Each assay was run at least in duplicate and the IC50 values were calculated using Life Science Workbench (LSW) Data Analysis Plugin for Microsoft Excel. The data were fitted into the formula y= Vmax / (1 + ([I]/IC50)). |
