| Description: | The 96 well plates were used, and the reaction volume was 25 μL using 50 mM sodium phosphate buffer (pH 7.0). For the screening, 30 nM of ADP-TAMRA with 2 μM SidA (based on the Bradford assay) and 20 μM of library compound were used, withthe final DMSO concentration at 2%. Fluorescence polarization measurements were performed with excitation at 544 nm and emission at 584 nm, using a wavelength cutoff of 570 nm. Anisotropy values were normalized to the values obtained in the negative control sample: SidA, ADP-TAMRA, and DMSO. The positive control consisted of ADP-TAMRA and DMSO. |
