Assay Name:  MAO-A Assay
Description:  Recombinant human MAO-A enzyme, expressed in baculovirus-infected insect cells, was purchased from Sigma-Aldrich (M7316). The assay was carried out in 96-well plates in a final volume of 200 μL at room temperature. According to the experiment protocol, a solution of test compound (2.0 μL) in DMSO (100%) was added to 90.0 μL of protein solution (0.3 μg prot/well, containing 6.6 μL protein and 9.993 μL phosphate buffer) and incubated for 30 min prior to the addition of 90 μL of freshly prepared Amplex Red reagent. The Amplex Red reagent was prepared as described for rat MAO-A. The enzymatic reaction was started by the addition of 20.0 μL/well of an aqueous solution of the substrate p-tyramine (150 μM final concentration). The p-tyramine solution was prepared from p-tyramine hydrochloride (45 μL) in water (100 mM) and phosphate buffer (2955 μL). Fluorescence measurements were performed for 45 min and the concentration-response curve of clorgyline (1.0 μL) serves as a positive control. DMSO (2.0 μL) was used as a negative control.
