| Fluorescence Polarization Assay |
Description: | In fluorescence polarization measurements, a fluorescent probe was excited with plane-polarized light and the degree of polarization remaining in the emitted light is measured. When the probe is bound to protein, it rotates slower than when free, and the remaining fluorescence polarization is higher. The fluorescent anisotropy value for a probe-protein mixture is the average of the value for bound and free probe. For the direct binding asaay, the data (anisotropy, A in mA vs galectin concentration, [G] in uM) were fitted to a simplified formula to obtain Kd. |
