| | ADA Enzyme Inhibition Assay |
| Description: | The activity of ADA was determined spectrophotometrically by monitoring the change in absorbance at 262 nm, due to the deamination of adenosine catalyzed by the enzyme. The change in adenosine concentration per min was determined using a Beckman DU-64 kinetics software program (Solf Pack TM Module). To correct for the background change in absorption, a reference blank containing all the assay components except the substrate was prepared. Each inhibitor concentration was tested in triplicate. The IC50 values were determined by linear regression analysis of dose-response curves. The Ki values were calculated from IC50 values by means of the Cheng and Prusoff. |
