| | ALR1 in vitro Inhibition Assay |
| Description: | The assay results were obtained at 340 nm and ALR1 inhibitory activity was measured with the absorbance change at this respective wavelength. Each well contains 200 無 of assay mixture containing potassium phosphate buffer 100 mM at pH 6.2 with test sample 20 無 of 10 mM, 70 無 enzyme preparation and 40 無 of10 mM sodium D-glucoronic acid as a substrate. This mixture was incubated at 37 蚓 for 5 min and 50 無 of 0.1 mM NADPH as a cofactor was added and the read was obtained at 340 nm. The assay mixture was incubated again at 37 蚓 for 10 min and the read was taken at the respective UV range in ELIZA plate reader. As positive and negative control, 10 mM valproic acid (20 無) and buffer solution (20 無) were used [Biol. Chem., 240:877-882]. |
