| | In vitro COX-1 and COX-2 inhibition assay |
| Description: | The reaction mixture contained reaction buffer solution (950 uL, 0.1 M Tris-HCl, pH 8.0 containing 5 mM EDTA and 2 mM phenol), COX-1 or COX-2 (10 uL), heme (10 uL), and test compounds (20uL). The reaction mixture was incubated for 10 min at 37 uC, followed by the addition of arachidonic acid solution(10 uL, making final concentration 100 uM) to initiate COX reactions. The COX reactions were stopped by addition of 1 M HCl (50 uL) after 2 min, followed by the addition of saturated stannous chloride (100 uL) and incubated for 5 min at room temperature. |
