| In vitro Urease Inhibition Assay |
Description: | Urease inhibition activity was determined by indophenol method. In brief, each 140 無 assay reaction contained 40 無 buffer (100 mM urea, 0.01 mM K2HPO4, 1 mM EDTA, and 0.01 mM LiCl2, pH 8.2), 10 無 of enzyme (5 U/mL), 10 無 of test compound, 40 無 each of phenol reagents (1%, w/v phenol and 0.005%, w/v sodium nitroprusside), and alkali reagent (0.5%, w/v NaOH and 0.1% active chloride NaOCl). Initially, urease enzyme and the test compound were incubated in the assay buffer for 30 minutes (in a linear reaction range) at 37 蚓, 40 無 of phenol and alkali reagents were added to stop the reaction. Enzyme assays were performed in 96-well plates, and a microplate reader was used to read the absorbance at 625 nm. All the assays were performed in triplicate. |
