| Description: | We screened an in-house library of 19 compounds using cis-FPP as substrate. Briefly, the condensation of IPP and GPP, FPP, or cis-FPP catalyzed by DPPS was monitored using a continuous spectrophotometric assay for diphosphate release in 96-well plates with 200 µL reaction mixtures containing 400 µM 2-amino-6-mercapto-7-methylpurine (MESG), 25 µM GPP, cis-FPP or trans-FPP, 200 µM IPP, 25 mM Tris-HCl (pH 7.5), 0.01% Triton-X-100, and 1 mM MgCl2. |
