| Comments: | Reactions were performed in methylacrylate cuvettes in 1 mL of reaction volume. The beta-lactamase assay contained 1 nM AmpC beta-lactamase, 200 uM Nitrocefin and varying amounts of Triton X-100 (either 0.00002% or 0.1%). Compound and enzyme were incubated for 5 minutes before the reaction was initiated by addition of substrate. Nitrocefin hydrolysis was monitored as an increase in absorption at 482 nm on an Agilent 8453 UV-visible spectrophotometer at room temperature. | |
