Assay Name:  Biochemical Assay
Description:  The potency of compounds to inhibit MALT-1 (Isoform1) enzyme was tested in a Fluorescent assay using recombinant MALT-1 (Isoform1, aa-840) generated in-house. The assay buffer was 50 mM HEPES (pH 7.5), 100 mM NaCl, 10 mM DTT, 1 mM EDTA, 0.9 M sodium citrate, 0.01% CHAPS. 1.5 nM of MALT-1 enzyme was incubated with various concentrations of test compounds (1 M) (1% DMSO) in the presence of buffer for 30 minutes at 30° C. The protease reaction was initiated by adding 50 μM of AC-LRSR-MAC (Peptide International, USA) substrate and incubated for 240 min. After 240 min incubation, fluorescence emission of the samples at 460 nm was measured at an excitation of 355 nm. The percent inhibition was calculated using the following formula: (Control GD−(Sample GD/Control GD))×100.
