| Description: | The AlphaScreen™ assay was performed following previous publication with minor modifications from the manufacturer's protocol (PerkinElmer, USA). A 20-μL reaction was set up in a PerkinElmer 384-well AlphaPlate where His-bromodomain at 10 nM was incubated with biotinylated JQ1 at 10 nM, nickel chelate acceptor beads at 12.5 g/mL, and tested compound at various concentrations for 15 min at room temperature, followed by the addition of streptavidin donor beads at 12.5 μg/mL and another 60-min incubation at room temperature. The plate was read on a Tecan Infinite M1000 Pro plate reader. |
