| FLIPR Assay |
Description: | The FLIPR protocol consists of 2 substance additions during a kinetic measurement. First the compounds to be tested (5 μM) are pipetted onto the cells and the Ca2+ influx is determined in comparison to the control (capsaicin 10 μM), providing the result as % activation and representing the compound-alone effect (calculation at peak signal related to baseline). A Ca2+ influx of 10%-60% reveals a partial agonist (pAG), a Ca2+ influx of >60% relates to a pure agonist (AG). After 5 min incubation, the Ca2+ influx is related to an injection of 100 nM of capsaicin and thereby the antagonistic effect of the test compounds detected. |
