| Description: | The substrate is linked to a fluorescent dye on one end and to a quenching group on its other end. The fluorescence of the substrate is significantly reduced due to intramolecular resonance energy transfer to the quenching group. Upon substrate cleavage by the enzyme, there is a disturbance of the energy transfer resulting in the enhancement of the fluorescent signal. The enzyme activity is determined by an increase in the relative fluorescent of reaction mixture, with umbilliferone excitation/emission filter set. |
