| Description: | A typical reaction mixture contains kynurenine, alpha-ketobutyrate, PLP and hKAT-I in the presence of test compound. The reaction mixture was incubated for 20 min at 38 deg C, and the reaction was stopped by adding an equal volume of 1 M formic acid. The product KYNA was detected by high-performance liquid chromatography and quantified on the basis of the absorbance at 330 nm as measured by a Hitachi L-7400 UV detector. The assays for each inhibitor were performed four times, and the data were analyzed using the SigmaPlot enzyme kinetics module (SPSS Inc.). |
