Assay Name:  β-Secretase (BACE-1)
Description:  β-Secretase (BACE-1, Sigma) inhibition studies were performed by employing a peptide mimicking APP sequence as substrate (methoxycoumarin-Ser-Glu-Val-Asn-Leu-Asp-Ala-Glu-Phe-Lys-dinitrophenyl, M-2420, Bachem). The following procedure was employed: 5 uL of test compound (or DMSO, if preparing a control well) were pre-incubated with 175 uL of the enzyme (in 20 mM sodium acetate containing CHAPS 0.1% w/v) for 1 h at room temperature. The substrate (3 uM, final concentration) was then added and left to react for 15 min. The fluorescence signal was read at λem=405 nm (λexc=320 nm). The DMSO concentration in the final mixture maintained below 5% (v/v) guaranteed no significant loss of enzyme activity. The fluorescence intensities with and without inhibitor were compared and the percent inhibition due to the presence of test compounds was calculated. The background signal was measured in control wells containing all the reagents, except BACE-1 and subtracted.
