Description: | Monkey: This assay was used to determine the inhibition effect of the present compounds on the activity of monkey GnRHr protein expressed by monkey GnRHr/CHO stably transfected cell lines.1. Experimental ProtocolA mammalian expression vector containing the monkey GnRHr gene was transferred into CHO cells by adding Lipofectamine LTX reagent containing Plus . Antibiotics were added the next day for screening to pick out the monoclonal cell lines.The monkey GnRHr/CHO stably transfected cell lines were inoculated in 96-well plates with an inoculation density of 25,000 cells/well. The culture medium was removed the next day, and loading buffer containing Fluo-4 dye was added to the plate (100 μL/well) and incubated for 30 minutes at 37° C. The plate was moved to room temperature and equilibrated for 10 minutes. Each compound was diluted with DMSO to seven concentration gradients of 100 μM, 10 μM, 1 μM, 0.1 μM, 0.01 μM, 0.001 μM, 0.0001 μM. Then, 1 μl of each gradient was added to each well and incubated for 10 minutes at room temperature. After automated addition of 50 L of GnRH polypeptide stimulant solution, the value was immediately detected at 494/516 nM by a microplate reader (flexstation 3). IC50 values of the compounds were calculated by software from different fluorescence signals at various corresponding concentrations. |
