Description: | Whole cell patch clamp recordings of hERG current (IhERG) were made at 37 °C from human embryonic kidney cells (HEK 293) cells stably expressing hERG (generously donated by Dr Craig January).15 Cells were maintained in culture and prepared for electrophysiological recording as described previously [Cheng et al., Brit. J. Pharmacol., 165:2260-2273; Du et al., J. Cardiovasc. Electrophysiol., 22:1163-1170]. Cells were superfused with a normal Tyrodes solution containing (in mM) 140 NaCl, 4 KCl, 2 CaCl2, 1 MgCl2, 10 Glucose and 5 HEPES (titrated to pH 7.45 with NaOH). Patch-pipettes were filled with a K+-based dialysate containing (in mM): 130 KCl, 1 MgCl2, 5 EGTA, 5 MgATP and 10 HEPES (titrated to pH 7.2 with KOH). Pipette resistance typically ranged from 1.53.5 MR and ∼70-80% series resistance could be compensated. |
