| Description: | The enzymatic reaction was performed in 384-well microplates with a final volume of 10 L containing 1% DMSO. Recombinant Human catalytic domains of MMP1 were assayed with the substrate according to supplier recommendation (ENZO life sciences) and after a preincubation with serial dilutions of tested inhibitors (from 10000 nM to 0.04 nM). After 10 minutes of incubation at room temperature, the fluorescence was measured using a microplate reader (320/425 nm). Data were normalized using the average of positive controls (enzyme with no inhibitor) and negative controls (no enzyme). IC50 were calculated using a four-parameters logistic model. |
