| Description: | DPP-VIII was obtained and purified. Using Gly-Pro-AFC as the substrate, DPP-IV inhibitors as set forth in Table 2 below were assayed for DPP-VIII inhibitory activity. Enzymatic reactions were conducted for various concentrations of DPP-IV inhibitors, using 0.0049 μg DPP-VIII and 100 μM Gly-Pro-AFC in a buffer solution containing 50 mM HEPES (pH 7.5) at room temperature. IC50 values of the inhibitors were determined by tracing the reaction rate using a fluorescence spectrometer for 60 min and analyzing the results using TableCurve 2D (v 5.01). DPP-IV inhibitors were analyzed using the fluorescence spectrometer at the excitation and emission wavelengths of 405 nm and 505 nm, respectively. |
