| Description: | The assay buffer contained 100 mM KHPO4 (pH 7.0), 2 mM EDTA and 2.5 mM reduced GSH. 100 µL of reaction buffer containing human recombinant mPGES-1 was added to 96-well non-binding plate, followed by the addition of test samples to their respective wells. The reaction mixture was incubated for 30 min at 20 °C. The reaction was initiated by adding 20 µL of cold 2.8 µM PGH2 to all wells and incubated again for 30 seconds at room temperature. The reaction was quenched by adding 20 µL of SnCl2 solution in 1 N HCl. |
